AAbstractEffects of Betaine substitution to methionine on BHMT expression in laying hens under heat stress Dr. Mahmood Nazari Department of Animal science, Faculty of Animal Science and Food Industries, Khuzestan Ramin Agriculture and Natural Resources University, Khuzestan, Iran Email: Fat_sa_2005@yahoo.com To investigate the effects of dietary betaine (Bet) substitution to methionine on hepatic betaine - homocysteine methyltransferase (BHMT) gene expression in laying hens under heat stress, an experiment with 144 Hy-line W-36 leghorn (at 50 to 62 weeks old) in complete randomized design (CRD) with a 3~2 factorial arrangement of treatments with eight treatments, 3 replicates and 8 hens in each replicate was performed. There were 3 dietary treatments: three doses of Bet (0, 13 and 26%) substitute to Met combined with two environmental conditions (thermoneutral (23‹C constant) and heat stress (23.9 to 35‹C cycling) conditions). After extraction total RNA from liver, quality was measured and was used to cDNA synthesis. Finally, BHMT gene expression was evaluated by real- time quantitative PCR (RT-qPCR). In this procedure, ƒÀ-actin was used as a house-keeping gene to normalize the gene expression data in the RT qPCR. The results indicated that performance parameters of laying hens (egg production and egg weight) and egg quality (eggshell strength, eggshell thickness) were all significantly reduced by heat stress (HS) (P<0.05). Substitution of methionine with Bet led to significantly increase of egg production (P<0.05), whereas it had no effect on egg weight and egg quality. However, BHMT gene expression was reduced 1.58 fold by heat stress when compared with control group. Moreover, BHMT gene expression was increased 2.11 fold by substitution of methionine with Bet. The results suggested that the increase in production concomitant with substitution of methionine with Bet probably due to increase in BHMT gene expression.
